Quantitative Analysis and Visualization Methods for Microgel Microscopy Data

Motivation

Microgels observed by single-molecule localisation microscopy appear as point clouds whose points correspond to the dye molecules. Within the G3N project we will develop computational tools for a comprehensive quantitative analysis of point cloud data, compute features that fully characterise a microgel, and measure and visualise changes in the soft and deformable microgels under different experimental conditions. Generative models will allow us to make predictions that can be tested experimentally. Combining point cloud data with pixel data from other modalities complements the computational toolkit and promises a more complete structural description of microgels.

Scientific Questions

Localisation-based fluorescence microscopy methods generate point clouds whose points correspond to the positions of individual dye molecules in the microgel. Algorithms are therefore being developed to extract features from these point clouds in order to describe different microgels. In addition, generative models are being developed that will ultimately enable a quantitative description of the shape and density of microgels under different experimental conditions.
